Descriptif du poste

The research team Pediatric Inflammatory Skin Disorders: Mechanisms and Therapeutic Implications, led by Dr. Elodie Bal at the Imagine Institute (Paris 15), is seeking a Postdoctoral Fellow. The selected candidate will focus on the genetic and pathophysiological study of severe pediatric forms of scleroderma.

Scleroderma is a rare, complex, and heterogeneous immune disease characterized by fibrosis of the skin and some internal organs, often leading to profound disability and reduced life expectancy. It is a multifactorial condition resulting from the complex interplay of genetic, epigenetic, and environmental factors.

In this project, our aim is to elucidate the underlying mechanisms of fibrosis in scleroderma using multi-omics approaches. The selected candidate will study the functional impact of candidate mutations on the fibrotic phenotype in various cellular models (2D and 3D). Our goal is to validate targets with significant diagnostic and therapeutic potential.

Our approach involves several key steps :

1. Molecular dissection of severe pediatric forms of scleroderma by integrating whole-genome sequencing and single-cell RNA sequencing to identify key genetic drivers and cells in the dermal microenvironment contributing to fibrosis ;

2. Functional characterization of genetic factors to elucidate the underlying mechanisms by which they promote fibrosis ;

3. Validation and modeling using cultured fibroblasts (patients-derived or CRISPR-modified cells) and a three-dimensional skin model which will also serve as a platform for future therapeutic screening.
